CRISPR/Cas9是一种通过Cas9/gRNA复合物对靶基因位点进行切割诱导DNA修复的方式实现特定基因修饰的基因编辑技术。Cas9/gRNA复合物在靶位点进行切割，产生双链断裂（DSB：Double-strand break），迫使细胞进行紧急修复。通过非同源末端连接（NHEJ：nonhomologous DNA end joining）可导致移码突变，实现基因敲除（KO）；通过同源重组修复（Homologous recombination, HR）可将外源片段整合到基因组指定位点（KI）。
