1921年C.C.Little用Abby Lathrop小鼠近亲培育数个近交系，使编号为57的雄鼠与52号雌鼠交配，培育成C57。其中毛色固定为黑色的培育成C57BL，BL是英文Black（黑色）的缩写。1937年，Little将维持的C57BL父系进行分离，将第10组亚系定名为C57BL/10。C57BL/10缺失等位基因Tlr4lps-del，导致mRNA和蛋白的缺失，因此对脂多糖的刺激表现为应答缺陷。
