使用基因编缉技术删除Rag2基因exons，建立Rag2基因敲除小鼠模型。对重组激活基因（Rag2）的整个蛋白编码区进行敲除。Rag2基因长度大约18kb，不产生成熟的T细胞或B细胞。Rag1和Rag2基因的编码区和3‘端的非翻译区包含在同一个单独内含子。对基因打靶小鼠动物模型的研究证明了Rag1和Rag2在V（D）J基因重排和淋巴系细胞发育中的关键性作用。rag1-/-和rag2 -/-小鼠有同样的表现型，严重的T/B细胞早期发育停滞，T细胞停滞于CD3-CD4-CD8-CD25+阶段，而B细胞停滞于B220- CD43+IgM-祖B细胞阶段。它们的外周血没有成熟的循环T/B淋巴细胞，这与人重症联合免疫缺陷综合征SCID非常相似。主要用于造血和免疫系统缺陷中的Rag2基因功能研究，癌症和毒理学研究（作为移植宿主）以及血液学，免疫学和炎性的研究。
