使用基因编缉技术删除Rag1基因exons，建立Rag1基因敲除小鼠模型。Rag1敲除小鼠T细胞和B细胞发育缺陷。胸腺、脾脏变小，无成熟T细胞、B细胞，NK细胞比例略增加。Rag1（Recombination-activating gene 1）是催化VDJ重组的酶复合物的组成部分 。Rag1敲除小鼠中没有成熟CD3+ TCRαβ+ T细胞，胸腺中细胞量比野生型或杂合子低15-130倍，胸腺细胞是CD8-CD4-且大多为IL2R+，脾脏及骨髓中检测不到IgM或IgD，显示成熟的B细胞完全缺失。
