使用基因编缉技术将人源的NTCP插入到小鼠NTCP位点上，破坏掉小鼠本身的NTCP，引入人源NTCP序列而建立人源NTCP小鼠模型。钠离子-牛磺胆酸共转运蛋白（Sodium Taurocholate Cotransporting Protein, NTCP）属于溶质运载蛋白家族，是一种钠离子依赖的牛磺胆酸转运蛋白，主要在在肝细胞表面表达，负责肝脏内的胆酸转运，胆汁酸排入小肠后，大部分被重吸收，通过NTCP被摄入肝细胞内，再次被分泌到胆汁中，形成胆汁酸的肝肠循环。2012年11月北京生命科学研究所（NIBS）李文辉团队研究发现，NTCP是一种主要的HBV/HDV功能性受体。NTCP表达水平的下调或沉默会降低HBV或HDV（带有HBV外膜蛋白）在易感细胞中的感染水平。发现NTCP扮演HBV侵染过程中的受体蛋白的角色解决了HBV研究领域的一个基础问题。目前越来越多治疗乙肝的新药把NTCP作为治愈乙肝的潜在靶标，比如Myrcludex B、环孢菌素A（Cyclosporin A）及其衍生物SCYX1454139，都是通过NTCP来阻断HBV进入肝细胞。而人（hNTCP）与小鼠（mNTCP）在157-165肽段具有极高同源性，但是由于84-87肽段的差异导致mNTCP仍然不能支持HBV/HDV的侵染，将84-87肽段置换为hNTCP的对应区段的mNTCP能够支持HepG2细胞被HBV/HDV感染。
